Oxidative stress regulating engineered polymers for the treatment of cancer and inflammatory diseases |

Reactive oxygen species (ROS) are a group of highly reactive small molecules generated by all aerobic organisms. The high level of ROS causes cellular damage and apoptotic cell death, depending on their concentration and induces inflammation diseases. Compared to normal cells, cancer cells are under oxidative stress associated with increased ROS production due to disrupted ROS homeostasis and utilize ROS to drive proliferation. Ironically, cancer cells have also adapted to oxidative stress by activating antioxidant systems. Therefore, cancer cells are susceptible to ROS insults induced by exogenous agents that weaken antioxidant defense or augment ROS generation to a level above which cells cannot survive. In this presentation, novel antioxidant and antioxidant polymeric prodrugs are discussed with emphases of rational design and proof-of-concept studies in animal models. |
