Engineering FcγRIIa for improved IgG Fc binding |
초록 |
Human Fc gamma receptors (hFcγRs) can modulate diverse antibody-dependent immune response via interaction with Fc region of IgG antibody. They are crucial to determine the efficacy in antibody-mediated cancer immunotherapy and are highly related with the initiation and progression of autoimmunity and inflammation as well. Due to the abundant expression on the surface of in a variety of leucocytes, FcγRIIa has been considered a critical target for various immunotherapies in spite of the low binding affinity to IgG antibody Fc. Using bacterial inner membrane display system, we have engineered FcγRIIa to improve IgG binding affinity and various strategies and potential applications for the resulting engineered FcγRIIa variants will be discussed. |
