제목 | Proteomic analysis of physiological properties of Candida magnoliae wild and its mutant strains |
초록 | Erythritol is a non-cariogenic, low-caloric (0.3 kcal/g) sweetener and safe for diabetics. The four-carbon sugar alcohol has about 70-80% the sweetness of sucrose and is found in fruits, mushrooms and some fermented foods The objective of this research includes identification and analysis of the expression patterns of proteins between wild and its mutant strains of Candida magnoliae which was able to produce erythritol with high yield. In an attempt to identify up-regulated or down-regulated proteins two-dimensional gel electrophoresis was performed for quantitative and qualitative comparison of total intracellular proteins. Most of the proteins were distributed in the range of pI 4-7 and MW 29-97 kD. Combination of 2-D PAGE and microsequencing allows identification of several of the overexpressed or low expressed proteins in the mutant C. magnoliae compared with the wild strain. As a result of identification, presumed proteins of citrate synthase, succinyl-coA synthetase, fumarase involved in TCA cycle, indole-3-pyruvate decarboxylase, and Yil144W cp were overexpressed in the mutant strain and Ygr 086 cp and enolase were over expressed in the wild strain. |
