Chitosan has found potential applications as a delivery carrier of siRNA, as chitosan has low toxicity, low immunogenicity, and positively charged amino groups which enable complex formation with negatively charged siRNA through electrostatic interactions. We prepared chitosan-based nanoparticles by complex formation with siRNA (mean diameter = 200 nm), and their physicochemical characteristics were investigated using electrophoresis, particle size analysis, and fluorescence correlation spectroscopy (FCS). The FCS technique was useful to understand the complex formation behavior between chitosan and siRNA, as well as to monitor the cellular uptake of the chitosan/siRNA nanoparticles in vitro. |
