We developed a novel shell cross-linked polymer micelle (SCM) that could preferentially release entrapped drugs in response to intracellular glutathione (GSH). The middle PLys shell of poly(ethylene glycol)-b-poly(L-lysine)-b-poly(L-phenylalanine) (PEG-b-PLys-b-PPha) copolymer micelles was cross-linked with a functional diamine cross-linker containing a GSH-cleavable disulfide bond. At an intracellular GSH level, the drug release was facilitated due to the efficient cleavage of disulfide cross-links. This new SCM may provide a platform technology for generation of useful intracellular nanocarriers of many poorly water-soluble drugs. Acknowledgement.
