제목 | Nano-/Micro- Encapsulation of Protein Drugs with Biodegradable Polymers via Electrified Coaxial Liquid Jets |
초록 | Production and control of particles of micrometer or even nanometer size with a narrow size distribution is of interest for many applications. These particles made of a small amount of a certain active pharmaceutical ingredient surrounded by a polymer are of particular importance for targeted drug delivery. The reasons of the interest in encapsulation are (1) to isolate an unstable component from an aggressive environment, (2) to avoid decomposition of an encapsulated component under a certain atmosphere, (3) to deliver a given substance to a particular receptor, and so forth. Among the methods to fabricate nano/micrometer sized capsules, the technique that uses electrohydrodynamic (EHD) forces to generate coaxial jets of immiscible liquids is recently highlighted.1 In this technique, high electrical potential is applied between ground electrode and two concentrically located needles where two immiscible liquids are fed into. For a certain range of values of the electrical potential and flow rates, a structured Taylor cone is formed at the exit of the needles with an outer meniscus surrounding the inner one.2 Small droplets or particles are formed as a result of the varicose break-up of the electrified jet. Herein, we fabricated nano-/micro- particles of protein drugs encapsulated via electrified coaxial liquid jets. To use these particles in drug delivery application, biodegradable and biocompatible polycaprolactone (Mn=10,000) was used as an encapsulating materials. The morphologies of fabricated nano-/micro- particles were studied using scanning electron microscopy (SEM) and the entrapment efficiencies were calculated by the bicinchoninic acid (BCA) protein assay using bovine serum albumin (BSA) as a model protein drug.
