Coaxial nanofibers with a unique core-shell structure can be served as drug delivery devices because encapsulated drugs in a core can be protected from harsh conditions. For coaxial electrospinning, a dual nozzle composed of an outer and an inner was employed to simultaneously electrospin both outer solution and inner solution. The outer solution was a mixture of PCL-PEG (1.5%, w/v) and PCL (13.5%, w/v) dissolved in MeOH:chloroform (1:1). For the inner solution, BSA was dissolved in 0, 5, and 10% PVA solution at a concentration of 10 mg/ml. Confocal microscopy revealed that fluorescent protein was successfully loaded in an inner core of coaxial nanofibers. Release studies were performed to examine dual release patterns of inner core proteins and surface-conjugated proteins in the coaxial nanofibers. According to concentration of PVA and inner/outer ratios, different release patterns of encapsulated proteins were observed. |
