제목 Inflammatory Proteinase Responsive Fluorogenic Hydrogel Lens Sensor
초록 Hydrogels have offered numerous biological applications such as implantable scaffold in tissue engineering, and biosensors due to their several advantages; (i) high water content enabling biological reactions taken inside the gel, (ii) high stability and biocompatibility and (iii) protection from non-specific adsorptions of proteins. Meanwhile, MMP2&9 have been investigated widely because they are highly related with inflammatory diseases. Specifically, cleavage of certain peptide sequences is accompanied during enzymatic reaction of MMP2&9. Herein, we developed MMP9 responsive lens-shaped fluorogenic hydrogel using peptide probe which specifically cleavable with MMP9. In detail, vinyl group terminated peptide probe was conjugated during formation of PEGDA hydrogel. Peptide probe inside the hydrogel sensor cleaved with existence of MMP9, becoming fluorescence off-state into on-state by FRET effect. Fluorescence intensity was increased with concentration of MMP9 increased.
