High-throughput single-molecule bioassay platform using single-point functionalized carbon-nanotube field-effect transistor arrays |
초록 |
Nanoscale field-effect transistor (FET) for biosensing has been emerged to a portable, manufacturable, and scalable bioassay platform. Despite the rapid progress on the fabrication of carbon nanotube FETs and in the manufacturing facilities, a critical challenge in bio-functionalization of the nanomaterials has been a major barrier to expand this technology to commercial biosensor application. Here, we show an advanced single-point functionalization chemistry that is electrically controllable at single reaction level and applicable to many devices in parallel in arrays. This single-molecule field-effect transistor (smFET) arrays are chemically identified by attaching single-stranded complementary DNA encoding DNA hybridization, an aptamer probe encoding small-molecule bindings such as neurotransmitters and bioaerosol toxins. This platform allows to monitor the single-molecule binding kinetics and temporally analyze the target concentration changes. |
