제목 Photo-induced Release of Plasmid from Crosslinked Nanoparticles
초록 While viruses efficiently package and protect their genome only to release their DNA or RNA at the appropriate stage of transduction, the engineering of triggered release in synthetic DNA particles has proven challenging since protection and release are competing design goals. We report the synthesis of o-nitrobenzyl and methacrylate functionalized polyethyleneimine for condensing plasmid (4.7 kbp) into particles as small as 100 nm after radical polymerization. DNA was not extractable from cross-linked nanoparticles until photoirradiated at 365 nm (3.5 mW/cm2) for 10 min. Plasmid condensed with uncrosslinked monomers transfected COS cells. However, crosslinked DNA nanoparticles produced no transgene expression until photoactivated, either extracellularly or intracellularly. These reagents may prove useful in the generation of stable DNA nanoparticles for temporally-controlled, spatial-addressed, and metered dosing of DNA for gene transfer.
