제목 3D vascular construction using engineered perfusable microfluidic devices
초록 Vascularization in three-dimensional (3D) environments is one of the crucial factors to be overcome in tissue engineering fields. Since cells are always placed at three-dimensional environments in living organisms, Optimization of vascularization in 3D environments has been attempted for many years. However, constructing cells and tissues over 500 um in thickness is pretty challenging due to lack of effective delivery of nutrient and oxygen to all tissue regions. Herein, we constructed a perfusable channeled device to fulfill the vascularized 3D tissue structure. The microfluidic device contains three perfusable channels (500 ㎛ diameter). We seeded human umbilical vein endothelial cells (HUVECs) in the channel, and the cells were fully proliferated to form endothelial monolayer in channel wall (~ 500 cells/㎟) with ≥70 % of cell viability for 2 weeks. Our perfusable vascular bed can be potentially used in large-size tissue regeneration as well as drug screening study.
