Acetate is an unwanted byproduct of microbial fermentation, but has a great potential as cost-effective and alternative carbon source in the production of various platform chemicals. This study demonstrates development and engineering of Escherichia coli strain for 3-hydroxypropionic acid (3-HP) production from acetate. Combinatorial strategies of metabolic engineering including the disruption of acetyl-CoA competing pathways, manipulation of glyoxylate shunt, improvement of cofactor (i.e., NADPH) supply through the TCA cycle, and redirection of the gluconeogenesis pathway have been employed.
