Vascular disease have still threaten human life until now. autograft has often been limited by the availability or shortage of donors. Therefore, there is an urgent demand for artificial grafts as substitutes. To overcome this problem, clinicians have used an industrially developed artificial vessel. However, this vessel has a problem of blocking a passage of the blood vessel induced by thrombus formation. To overcome this limitation, heparin has been studied. However, heparin can still cause heparin-induced thrombocytopenia (HIT) in blood. To reduce this side effect, we have developed covalently conjugated heparin and substance P to the Poly(l-lactide-co-ε-caprolactone). We could quantified how much heparin is attached to the PLCL through the toluidine blue assay and confirm whether artificial blood vessel is coated through SEM and TBA. Additionally, we investigated the effects of heparin and substance P on cell growth through in vivo test. |
