Synthetic biology is the study to design and construct new biological parts, devices and systems or re-design natural biological systems for useful purposes. When using genetic devices for many applications, the operational range and dynamic range should be adjusted in the dose-response curve. Therefore, we constructed riboswitch-based genetic devices with fast and easy methods without any information. First, naringenin riboswitches developed by evolutionary approaches. The naringenin riboswitch library were constructed and screened by tetA gene. As a result, we found six different naringenin riboswitches and validated them. Next, genetic circuits with a coenzyme B12 riboswitch were constructed for modular tuning of existing riboswitches. Two transcriptional repressor sets were used and various strength of constitutive promoters were used to regulate expression of riboswitch and the repressor gene. As a result, the circuit variants showed different dose-response curves. |
