Identification of 3-hydroxypropionic acid transporters and their application in Pseudomonas denitrificans |

3-Hydroxypropanoic acid (3-HP) is an important platform chemical which has been successfully produced in Escherichia coli, Saccharomyces cerevisiae, Klebsiella pneumoniae and Pseudomonas denitrificans. However, for commercial production of 3-HP more studies are still needed, including the one on 3-HP transporters. In this study, for the first time, we have identified 3-HP transporters and examined their effects on 3-HP production with glycerol as carbon source. Two promising candidate proteins, an exporter and an importer, respectively, were identified and several mutant strains with deletion of the importer and/or overexpression of exporter were developed. The mutant strains showed improved cell growth and 3-HP production. |
