A MALDI-MS-based rapid enzyme screening platform |

As the field of biotechnology becomes diverse and the demand for related technologies increases, researches on enzymes are actively being carried out. However, due to the nature of enzymes that selectively react with substrates, it is important to rapidly screen enzymes that required for a particular industry. Conventional enzyme screening methods are time-consuming and inconvenient because each substrate must be treated with various enzymes to individually check whether each reaction has proceeded. In this study, we proposed a rapid multi-analysis platform based on mass-assisted laser desorption/ionization mass spectrometry (MALDI-MS) to solve the inefficiency of enzyme activity screening mentioned above. Glucan, a polysaccharide consisting of glucose monomers, was used as a target substrate to demonstrate the concepts presented in this study. The MALDI-MS confirmed the activity of hydrolyzed enzymes that were specifically recognized by their glycosidic bond types and were able to be screened more effectively than conventional enzyme activity assays. |
