Osteoarthritis (OA) is a chronic inflamed joint disease and in OA inflammatory cytokines are higher than normal arthritis. Nuclear factor kappa B (NF-κB) signaling is the major pathway mediating the expression of proinflammatory cytokines. Therefore, NF-kB signailing is potential targets for arthritis treatment. In this study, we have developed of poly (lactic acid) (PLA)-hyaluronic acid (HA) di-block copolymers to deliver melittin safely to arthritis inflammation sites. Melittin has the potential as an anti-inflammation drug by inhibiting NF-κB activation, but the cytotoxicity of melittin has been attributed to both cell necrotic and apoptotic cell. To overcome this problem, we loaded melittin in to polymersome which is a biologically stable drug delivery system. Our results show that the MEL-PL system has high potential as a novel carrier for anti-inflammation treatment for arthritis by penetrating cell membrane directly and blocking NF-κB signaling in high efficiency. |
