제목 Enhancement of the PET-degradation potential of PETase through carbohydrate-binding module and surface expression system
초록 PETase is an enzyme that degrades Polyethylene terephthalate (PET), a potential environmental contaminant and pollutant. The conventional way to remove PET from water involves its physical entrapment followed by filtration through membranes, posing a limitation that particles of smaller size are not retained, and still viable. On the flip side, the enzyme PETase presents a more biologically environment-friendly way to degrade PET into monomers, providing a fundamental solution to the remediation of the microplastic-induced bioburden. Herein, the PETase was expressed on the surface of E. coli with the Carbohydrate-Binding Module(CBM) that binds to PET. The degree of surface expression over culture time and PET degradation ability were compared and evaluated with those in which only PETase was expressed on the surface of E. coli. The surface expression system addressed the shortcomings of the enzyme in terms of productivity, stability, and CBM-induced performance.
