Nano-biohybrid pH/temperature-sensitive hydrogel system for controlled release of human Growth Hormone |

In this study, we developed a nano-biohybrid system that combined of a novel pH/temperature sensitive copolymer hydrogel matrix and Layered Double Hydroxide (LDH) nanoclay particles for controlled release of human Growth Hormone (hGH). The hydrogel matrix is contained of temperature-sensitive triblock poly(e-caprolactone-co-lactide) - poly(ethylene glycol) - poly(e-caprolactone-co-lactide) (PCLA-PEG-PCLA) and pH-sensitive cationic moiety poly(amino ester urethane) (PAEU). The synthesis of hydrogel was well confirmed by H-NMR and GPC. XRD characterization was used to affirm the intercalation in nano-biohybrid system. Their mechanical strength was examined by rheological viscometer. Sol-gel phase diagram indicated the pH/temperature sensitivity of the system. Furthermore, this system can form gel rapidly after injecting subcutaneously into male SD rats. The in vitro and in vivo stability were checked to satisfy the demand of sustained delivery system. The result of in vitro and in vivo release of hGH demonstrated that the addition of LDH nanoclay can prolong the release period in comparison with only hydrogel system. |
