In this study, we propose intracameral delivery of azidobenzoic acid-modified chitosan (Az-CH) to develop a rat glaucoma model. For this, we pursued to chronically elevate the intraocular pressure(IOP) and thus, after intracameral injection of Az-CH in rats, we applied UV irradiation to photo-crosslink the Az-CH in the eye. To test this approach, we employed 24 eyes from 12 rats, which were divided into six distinct groups: 2 mg/ml Az-CH with UV, 10 mg/ml Az-CH with UV, 2 mg/ml Az-CH without UV, 10 mg/ml Az-CH without UV, no Az-CH with UV and normal control. After treatment, the change in IOP was monitored weekly, using a TonoLab® tonometer. The best result in IOP elevation was observed with the 10 mg/ml Az-CH with UV irradiation group (218.0%), which was maintained for up to 3 weeks after treatment. Therefore, we conclude that intracameral delivery of Az-CH, followed by photocrosslinking via UV irradiation, can effectively induce chronic IOP elevation to develop a rat glaucoma model. |
