제목 | Maskless Photolithography System using Micromirror Array for Peptide Array Synthesis on a Chip |
초록 | We report a maskless photolithography system using a micromirror array that can be used to synthesize protein ligands and peptides as a microarray on a chip via photosynthesis. The computer-controlled peptide synthesis system developed can build a number of different peptide molecules on a chip, and is an efficient method to quantify the different binding activities of peptides on a chip. The assay time to obtain more precise binding assay results between protein and peptide ligand is remarkably reduced on a chip compared to a conventional peptide synthetic method using bead resins, because parallel synthesis of protein ligands or peptides and parallel affinity assays are possible using a chip. HPQ-containing pentapeptides were built up on a slide glass by the photosynthesis chemistry using NVOC(nitroveratryloxycarbonyl)) protecting group in the automated synthesis system as a model experiment. The binding assay between the synthesized pentapeptide ligands and streptavidin was performed using the peptide array chip since HPQ-containing pentapeptides are well known to bind streptavidin. Compared with a conventional peptide synthesis experiment using bead resins, assay time was remarkably reduced on a chip within a day from one week to obtain the same assay results. Consumed reagents volume and assay cost were also remarkably saved with the help of peptide chip. We expect that the automated peptide synthesis systems can build up a number of different peptide molecules on a chip and give efficient methods to quantify different binding activities of peptides on a chip. |
