제목 Hydrogel scaffold enabled with stem cell and growth factor delivery for enhanced bone regeneration by paracrine effects
초록 This study investigated paracrine effect between mesenchymal stem cells (MSCs) and osteoblasts in the biopolymer-based three-dimensional scaffold. To create indirect co-culture system, osteoblasts were encapsulated in alginate microbeads using electrostatic droplet extrusion. They were co-encapsulated with MSCs in biopolymer bulk hydrogel scaffold. We hypothesized osteoblasts enhances osteogenesis in MSCs by the paracrine effect. Therefore, we studied three experimental groups, osteoblasts with MSCs, bone morphogenetic protein 2 (BMP-2) with MSCs and only MSCs in 3D hydrogel culture to compare the efficiency of paracrine effect. Our system will show the advantage to study osteogenesis in MSCs by separating MSCs in bulk hydrogel and alginate encapsulated osteoblasts.
