제목 Cellular Engineering of Insulin-Secreting Pancreatic Islets Using Mussel Power
초록 For successful islet transplantation, main issues are protection of transplanted islets from host’s immune system and hypoxic state in large size of islets. To carry out surface modification of pancreatic islets for protecting them from immune reactions, branch-type poly(ethylene glycol) was modified to catechol moiety having mussel power and grafted onto islet surface. The viability and functionality of PEG-catechol-grafted islets were evaluated. On the other hand, polydopamine-micropatterned poly(dimethylsiloxane) film was used for generating optimal size of islets. Medium droplet containing dissociated islet single cells was dripped on the film, and it was turn upside down to hang the droplets. By this environment, the single cells were aggregated by the micro-gravity in the hanging droplet. We could control size of islet-like spheroids with different cell number in each droplet. Based on these findings, we can use the mussel-inspired materials for successful islet transplantation.
