Micropatterning Technique-Based Immunoprotective Device to Transplanted Pancreatic Islets for Diabetes Therapy |
초록 |
Pancreatic islet transplantation is a tremendous strategy for curing diabetes mellitus because transplanted islets can actively recognize the dynamic fluctuation of blood glucose level in blood stream. However, transplanted islets are rejected from the diabetic recipients via host immune reaction and hypoxic mechanism in the transplantation site. To fight out these limitations, we are currently focused on the micropatterning that has become a standard in biomaterials engineering. Here polydimethylsiloxane-based the embossing sheet and microfluidic chip were fabricated and then live islet cells were encapsulated in them. These devices protected islets from immune reactions, while their viability and functionality were well maintained. These results indicated that the device having pancreatic islets dynamically managed the blood glucose level in diabetic mice. Therefore, this micropatterning based immunoprotective device could be utilized for successful pancreatic islet transplantation. |
