The effect of reactive oxygen species (ROS) on cell fate depends on the level of ROS presented. High levels of ROS cause detrimental oxidative stress and can lead to cell death. However, low levels of ROS provide a beneficial effect, accelerating cell proliferation. Therefore, we fabricated nerve conduits containing ROS generator using cellulose acetate. We choose protoporphyrin IX (PpIX) as a ROS generator, which was grafted onto the surface of cellulose monolith (CA-PpIX). We determined the amount of ROS grafted on the surface of CA-PpIX. Schwann cells were incubated with various concentrations of PpIX and laser intensity to measure the optimal concentration of the ROS. As a result, the degree of differentiation of schwann cell was different according to amount of PpIX grafted and various laser irradiation. We have found optimal conditions for cell proliferation with CA-PpIX. Our results show that the CA-PpIX may be a good candidate for enhancing nerve regeneration. |
