제목 Single-Cell Level Observation of Cell Penetrating Peptide-mediated Endocytosis Using Plasmonic Nanoprobes
초록 Cell penetrating peptides (CPPs) can efficiently deliver membrane-impermeable cargoes (i.e., proteins, nucleic acids, drugs, and etc.) into the cytosol of cells, which have drawn much attention in biomedical fields. However, the behavior of intracellular CPP is poorly understood. Herein, we propose a real-time monitoring and imaging method for CPP-mediated endocytosis using plasmonic gold nanoparticles (GNPs) as imaging agents. The CPP-conjugated GNPs were systematically characterized in terms of size, zeta potential, absorbance, and scattering properties. Using the optimized CPP-GNPs, we evaluated the efficiency of cellular uptake mediated by CPP-conjugation using a dark-field microscopy-based hyperspectral imaging system. Also, by measuring the dynamic changes in scattering properties of the probes, it was possible to monitor the cellular uptake and sequential endosomal escape in the cytosol. The plasmonic gold nanoprobe-based imaging method provides an advanced method to predict the mechanism of intracellular CPP- mediated endocytosis of cargoes.
