Synthesis of Ni2+-NTA –polystyrene nanoparticle encapsulated Fe3O4 magnetic particle as an efficient tool for His-tag protein purification |

An efficient and simple strategy to purify recombinant proteins has been needed with increase of the various recombinant proteins for diverse applications. Currently, the method of giving affinity tags, eg. Histidine tag, to proteins has been commonly used to facilitate the protein separation. In this work, we developed Ni2+-NTA-surface functionalized polystyrene (Ni2+-NTA-PS) nanoparticle encapsulated with Fe3O4 nanoparticles for affinity purification of His-tagged recombinant proteins. As the Ni2+-NTA group allows immobilization of His-tagged proteins, the Ni2+-NTA-PS particle containing magnetic Fe3O4 nanoparticles can separate proteins by magnet. The Ni2+-NTA-PS was dissolved with Fe3O4 nanoparticles in chloroform and ultra-sonicated in aqueous phase to form mini emulsion. After solvent evaporation, the particle was characterized by DLS, TEM and SEM measurement. The separation of the His-tagged proteins from lysate was conducted by using magnetic property of this particle. |
