제목 Synthesis of MethoxyPoly(ethyleneglycol)/Polyesters Diblock Copolymers and Evaluation of micellar characterization as drug carrier
초록 The development of various biomaterials containing poly(ethylene glycol) (PEG) has been demanded for biomedical research. In the field of drug delivery, PEG as a drug carrier possesses great advantages, such as easy drug loading and releasing, long circulation after intravenous administration, and easy penetration of natural barriers such as the nasal mucosa. However, the problem of nonbiodegradability has been raised while the use of materials containing PEG has been extended. The endowment of biodegradability can be achieved through the conjugation of biodegradable segments onto PEG.
In this study, diblock copolymers consisting of methoxypoly(ethyleneglycol) (MPEG) and poly(ε-caprolactone) (PCL), poly(δ-valerolactone) (PVL), poly(L-lactic acid) (PLLA), or poly(lactic-co-glycolic acid) (PLGA) as biodegradable polyesters were prepared to compare the characterization of diblock copolymers. MPEG-PCL and MPEG-PVL diblock copolymers and MPEG-PLLA and MPEG-PLGA diblock copolymers were synthesized by the ring-opening polymerization of ε-caprolactone or δ-valerolactone in the presence of HCl · Et2O as a monomer activator at room temperature and by the ring-opening polymerization of L-lactide or a mixture of L-lactide and glycolide in the presence of stannous octoate at 130 oC, respectively. The synthesized diblock copolymers were characterized with 1H NMR, GPC, DSC, and XRD. The micellar characterization of MPEG-Polyester diblock copolymers in an aqueous phase was carried out by using NMR, dynamic light scattering, AFM, and fluorescence techniques. Most micelles exhibited a spherical shape in AFM.
In conclusion, we investigated that characterization of micelles showed difference of thermal properties, crystallinity, critical micelle concentration, and diameter of micelles dependence on the polyester block segments. Also, we confirmed that the micelles formed with MPEG-Polyester diblock copolymers have possibility as a potential hydrophobic drug delivery vehicle because a hydrophobic drug could be preferentially distributed in the micelle core.
