Polymer hydrogels are three dimensional polymer networks containing a large amount of water. Due to their ability to simulate the nature of most soft tissues, hydrogels are a highly attractive material for developing synthetic extracellular matrix (ECM) analogs. Matrigel is a kind of hydrogel which is generally used for basement membrane matrix of stem cells. It consist of laminin, collagen IV, entactin, and growth factors. Unfortunately, relatively little is known of the growth requirements for neural differentiation in matrigel. In the present study, we show that some ECMs supports the generation of 3D brain tissue, so-called cerebral organoids, which closely mimics the endogenous developmental program. In this study, we encapsulate embryoid bodies generated from hPSCs into ECM hydrogel. The environment factors which affect neuron morphology of cell are investigated. |
