In this study, twenty kinds of kojic acid-tripeptide amide library was prepared by solid-phase Fmoc/tBu strategy using Rink Amide SURE® resin. To synthesize kojic acid-tripeptide amides, the reaction condition was optimized to minimize time. Prepared kojic acid-tripeptide amides were compared their inhibition activity to kojic acid-tripeptide free acids. Then, we confirmed the position effect of amino acid in tripeptide coupled to kojic acid. At last, kojic acid-tripeptide amides which showed high inhibitory activity, were compared their inhibitory activity and stability to kojic acid and kojic acid-tripeptides free acids by mushroom tyrosinase inhibition assay. As a consequence, the peptide sequence of FWY coupled to kojic acid! has high inhibitory activity and maintained that with changed time, temperature and pH value. |
