Chondrocyte Culture Using a Nanofiber-Hydrogel Composite System |

Cartilage is a flexible connective tissue covering the end part of bones in joints. Cartilage has a limited capacity for self-repair. Therefore, tissue engineering approach is necessary for the treatment. In this study, a nanofiber-hydrogel composite system has been investigated as a cell and growth factor carrier for cartilage tissue engineering applications. Rabbit articular chondrocytes and transforming growth factor-β1 (TGF-β1) were mixed with the precursors of a heparin-based hydrogel. The mixture was then seeded onto a roll-type scaffold made of electrospun PLCL/gelatin fibers. The nanofiber-hydrogel composite system showed more improved cell seeding efficiency and enhanced chondrocyte-specific gene expression by real-time RT-PCR than those prepared without hydrogel. Results indicate the potential of the PLCL nanofiber-heparin hydrogel composite system for cartilage tissue engineering.
