Early diagnostic system for breast cancer, 2nd cause of cancer death in women is a key to effective treatment. To realize this purpose, an ultrasensitive electrochemical aptasensor for multiplexed detection of exosomes using different signals of metal ions was developed. Specifically, carbon electrode modified with AgNPs and other composites was employed to immobilize CD63 aptamer to capture exosomes. Once captured onto electrode, EpCAM and/or HER2(+) exosomes bind with their specific aptamers containing primer region and RCA is initiated by primer to produce repeats of poly G and T, which consequently forms complex with copper and lead ions, respectively. The proposed diagnostics for multiplex exosome analysis system successfully achieved a detection of limt (LOD) at 1 particle/ml with high specificity and could be a clinical platform to diagnose breast cancer via liquid biopsy. |
