제목 Near Infrared Dye-Conjugated oxidative stress generating polymeric micelles
초록 Chemotherapy has several inevitable limitations such as severe side effect. One promising strategy to enhance the therapeutic efficacy is the combination with other treatment modality such as photothermal therapy(PTT). In this work, we developed IR820-conjugated Reactive Oxygen Stress generating polymeric micelles(HRGP-IR) as a dual imaging agent and a combinational anticancer therapeutic agent. HRGP was synthesized from Michael addition polymerization and HRGP-IR was then prepared by conjugating IR820. HRGP-IR was designed to self-assemble to form thermodynamically stable micelles. In cancer cells, HRGP-IR micelles could generate hydroxyl radical by fenton reaction and induce apoptosis in combination with NIR laser irradiation. In mouse xenograft model, HRGP-IR micelles exhibited significant fluorescence and photoacoustic signal in tumor, and upon NIR laser irradiation, the temperature of tumor of HRGP-IR micelles treated mice was significantly elevated and tumor were eradicated.
