Branched-PEI conjugated Cationic Polymer for Gene Delivery with Low Cytotoxicity and High Transfection Efficiency |

The aim of this study was to investigate the effect of branched-PEI(bPEI) conjugated cationic polymer such as chitosan on in vitro effect of transfection efficiency and cytotoxicity at varying cell lines. O-carboxymethyl chitosan(CMCh) was prepared by chemical reaction chitosan with mono-chloroacetic acid to C6-OH posion of low molecular weight water soluble chitosan(LMWSC). MTT assay indicated that OCMPEI complex was significantly less toxic than bPEI. The transfection efficiency in cancer cell and normal cell of the OCMPEI copolymers were evaluated. Results of transfection efficiency showed that the highest transfection efficiency was observed in OCMPEI complex with pDNA or siRNA(16:1,w/w). It is expected that the OCMPEI complex may be a good carrier for gene delivery.
