Gene therapy is a sort of remedy by delivering therapeutic gene into target cells. However, the therapeutic gene has short half-life and low stability due to its denaturation. To overcome the limitation, branched polyethyleneimine carbon dots (bPEI CDs) and human mesenchymal stem cells (hMSCs) were used as a carrier of therapeutic gene. The bPEI CDs are carrier for delivery therapeutic gene into hMSCs. Synthesis of bPEI CDs was conducted with a green, facile pyrolysis using microwave. The bPEI CDs have low cytotoxicity and facilitate bioimaging after gene delivery. The raw bPEI indicated cytotoxicity, but bPEI CDs presented low cytotoxicity levels at all tested concentrations. The bPEI CDs could transfer therapeutic gene more safely and could also induce cellular fluorescence imaging. The amount of secreted therapeutic protein from bPEI CDs/gene@hMSCs was higher than that from bPEI/gene@hMSCs. Consequently, toxicity alleviated bPEI CDs can be potential gene carrier for cancer therapy. |
