Development of pH- and thermo- sensitive hydrogel composite as injectable filler for vertebrae stabilization |

The study contains the fabrication and characterization of a hydrogel composite system with a chitosan polymer derivative and biphasic calcium phosphate (BCP) as bioactive materials. The system comprises of the gellable polymer and calcium phosphate source mixed with the forming solution. The injectable system readily forms at physiologic conditions (pH 7.0, 37oC). BCP particles were dispersed in the liquid phase solution using a dispersing agent. Morphological assessment both in vitro and in vivo was conducted to optimize setting conditions and properties of the injectable filler. The optimized conditions were used for the development of the injectable hydrogel composite for use as orthopaedic bone filling material specifically for vertebrae stabilization. Good cell-material interaction was observed for the samples based on the attachment and proliferation of cells in vitro determined via viability assay and confocal microscopy images. Implantation on rabbit femoral epiphysis of injectable hydrogel composite show no immune reaction as seen in histological images after one week implantation and new bone formation was observed after 4 weeks of implantation. The hydrogel composite showed great potential for use as an injectable filler for stabilizing vertebral fractures. |
