Preparation of Urease-Embedded Porous PDMS Membranes and Electrochemical Detection of Urea |

Bioelectroanalytical devices combining the specificity of enzymatic reactions with the high sensitivity of electrochemical signal transduction are important to real-time monitoring. In the current work, we successfully modify glassy carbon electrodes (GCE) with amine groups (−NH2) to produce aminated GCE that are capable of effectively detecting current changes during the urease-catalyzed decomposition of urea. We have investigated the use of porous PDMS as an enzyme immobilization medium. Such porous systems are suitable for enzyme embedding while the pores are generated in a reverse micro-emulsion system. A basic reverse microemulsion is prepared from a mixture of Brij35, cyclohexane and 1-hexanol. The urease-embedded porous PDMS membrane shows a high sensitivity of 200 µA mM-1 cm-2 and a linear dependence of the current on the urea concentration in the range of 0.2-1.0 mM, when employed for cyclic voltammetry urea sensing. |
