In Vitro and In Vivo Study of Bone Defect Formation Using a Novel Scaffold Containing Solid Free-form PLGA Microspheres |

Poly(lactice-co-glycolide acid) (PLGA) microspheres have been studied as drug delivery system aiming apply for bone tissue regeneration in previous work. The aim of this study continuously develops a novel scaffold containing solid free-form PLGA microspheres to examine in vitro and in vivo studies. For this purpose, two groups of scaffold were fabricated by solid free-form adhesion using the W/O/W1/W2 emulsion droplet formation method including scaffolds unloaded and loaded with simvatatin (SIM) drug. After obtain two kinds of scaffold, the morphology, cytotoxicity and cell proliferation on scaffolds were examined and compared. With the higher of SIM loaded solid-form adhesion PLGA (SIM-SFA PLGA) scaffold, the SIM release and scaffold biodegradable characteristic were then investigated. Furthermore, the confocal images of MG-63 osteoblast cells adhesion, growth and proliferation showed the excellent bio-activity of SIM-SFA PLGA scaffold. Beside, the in vivo study and history analysis data from the implantation of scaffold on femur rapid showed that the bone almost regenerated in the large defects after 8 weeks of implantation. Overall of this study revealed that SIM-SFA PLGA scaffold keep a promise for using in the clinical in the future. |
