Fabrication of Simvastatin Conjugated Porous PCL/BCP Composite Microspheres as Tissue Engineering Scaffold |

In this study, a scaffold fabrication method utilizing composite of biodegradable polymer PCL and bioactive ceramic BCP powder to create novel scaffold structure is presented. The characterization of PCL/BCP composite microspheres scaffold was investigated by scanning electron microscopy (SEM), micro-computed tomography (micro-CT), X-ray diffraction (XRD), and energy-dispersive spectroscopy (EDS). The morphology observation and porosity investigation revealed the contingent conjugation of microsphere to create new scaffold structure with distribution of pores in range of 0.02-500 µm. Both PCL and BCP components were homogenously distributed, and a high porosity was observed in the scaffold. Otherwise, the mechanical testing also showed the high mechanical yield stress of scaffold. In vitro study showed the attachment and spreading of osteoblast cells on surface of porous particle of scaffold after 1, 3, and 5 days of cell culture. The sustained release of simvastatin drug induced for prolonged period of osteoblast cell attachment and growth on scaffold. The results indicated that simvastatin conjugated porous PCL/BCP composite microspheres had good possibility to apply in tissue engineering. |
