Co-encapsulation of magnetic Fe3O4 nanoparticles and paclitaxel into electrosprayed PLGA microspheres for intratumoral drug delivery |

We fabricated magnetic (Fe3O4) PLGA microspheres (MPMs) containing Paclitaxel (PTX), an anticancer drug, of mean diameter 5±2µm using electrospraying method in order to develop more efficient drug delivery for cancer treatment. The microspheres were dense, well individualized and homogenous in size. The values of magnetizations for the MPMs showed good magnetic field intensity and super paramagnetic property. The PTX-MPMs showed high entrapment efficacy and supported a sustained and steady release of PTX. In vitro the PTX-MPMs were easily internalized into murine Lewis lung carcinoma cells and they induced apoptosis. In vivo, the PTX-MNPs showed higher anti tumor activity than free DOX solution. The antitumor activity of the PTX-MNPs was found higher with than without an external magnetic field in vivo. From the results of the magnetic PLGA microspheres may provide a therapeutic benefit by delivering drugs efficiently to magnetically targeted tumor tissues, thus achieving safe and successful anti tumor effects with low toxicity. |
