Efficient expression system for the production of heterologous polyketides in Streptomyces venezuelae |

The polyketide synthase (PKS) gene cluster responsible for tylactone biosynthesis from Streptomyces fradiae was expressed in a heterologous host, S. venezuelae. We constructed the engineered S. venezuelae mutant in which the PKS related to the biosynthesis of the native polyketides is deleted, and developed a two-vector system for heterologous expression of the five genes comprising the tylosin PKS gene cluster. Interestingly, the S. venezuelae transformants produced not only tylactone but its glycosylated derivatives, desoaminyl tylactone. The deoxysugar desosamine is synthesized by the products of desosamine biosynthetic genes in S. venezuelae. This result demonstrates that the glycosyltransferase (Des VII) is aglycone-flexible and attaches desosamine to tylactone, a 16-membered ring macrolactone. |
