제목 | Monitoring Hyaluronidase Activity Using Reducible Hyaluronic Acid Nanogels |
초록 | Introduction Indocyanine green (ICG), a near-infrared (NIR) fluorescent dye, has been clinically used as a superior and biocompatible probe for biopsy of sentinel lymph nodes. However, despite its great potential as a diagnostic probe, its application is still limited because of several innate limitations like poor physicochemical stability and low detection sensitivity. In this study, we present shell-crosslinked HA nanogels (sc-nanogels) generated via a reducible covalent linkage which incorporate ICG derivative for selective imaging of HAdases with appreciable signal-to-noise (S/N) ratios in serum and in vivo. Method sc-nanogels with ICG were fabricated via both non-covalent interactions and cleavable chemical bonds for significantly increased specificity of probes as well as an enhanced physicochemical stability in serum conditions Result An ICG fluorescence signal of sc-nanogels was specifically switched on only in the presence of a hyaluronidase, an enzyme for the cleavage of HA. In addition, sc-nanogels exhibited long-term stability in aqueous solution at room temperature, compared to common ICG. Conclusion Sc-nanogels might be facilitated for long-lasting and selective imaging and tracking for lymph nodes, cancer cells, and therapeutic cells in vivo. |
