Optimizing Culture Medium for Cell Growth of Chinese Hamster Ovary Using Design of Experiments (DoE) |

The Chinese hamster ovary (CHO) cell has been used extensively as the preferred mammalian cell host for the production of recombinant proteins. Among the multiple benefits on using this system, CHO cell can achieve the appropriate post-translational modification and produce recombinant proteins with human-compatible glycosylation. Recently, with the increase in demand for therapeutic proteins, many studies have been performed to improve protein production in CHO cell. In this study, we optimized the culture medium of CHO cell for the enhancement of cell growth. First, we cultivated CHO cell using commercial media to select a suitable culture medium. Then, the Plackett-Burman design (PBD) was used to evaluate the effect of various 29 supplementations including hormones, vitamins, and lipids on cell growth. The PBD results showed that the most effective components for cell growth of CHO cell were two vitamins. The concentration of the major components was optimized via response surface methodology (RSM) with a central composite design. The RSM results showed the increased maximum viable cell density compared to the non-supplemented medium. |
