In this study, we used antisense oligodeoxinucleotides (ODN) and small interfering RNA (siRNA: RNA duplex that is 21 to 23 base pairs long) expression vector system to block the expression of green fluorescence protein (GFP) gene within target cancer cells., Antisense ODN and siRNA mediate strong and specific suppression of targeted gene. However, target tissue specific delivery of these oligos is one of the major hurdles to their therapeutic applications. To overcome this limitation, we first synthesized cationic polymeric conjugate, poly (ethyleneimine)-poly(ethylene glycol)-folic acid (PEI-PEG-FOL) and used as a targeted delivery carrier for a anti-GFP siRNA expression vector (pSUPER-siGFP). Antisense ODN specific for GFP gene was conjugated with PEI-FOL for targeting cancer cells and then produced polyelectrolyte complex with lipofectamine. These polyelectrolyte complexes showed a significant decrease in the expression level of the GFP gene in target cancer cells. |
