The cell-free therapeutics to foreignize tumor cells with non-self antigens has received huge attention as a mean to induce cytotoxic T lymphocyte (CTL)-mediated immunological rejection of tumor cells. In this study, we prepared matrix metalloproteinase 9 (MMP9)-sensitive polymeric conjugates bearing PEGylated hyaluronic acid (HA) as the tumor targeting moiety and ovalbumin (OVA) as the model foreign antigen. The MMP9-sensitive peptide linker was chemically attached between PEG and the HA backbone to promote the removal of the PEG from the polymeric conjugates at the tumor tissue. When the conjugates were systemically administered into the tumor-bearing mice pre-treated to have OVA-specific CTLs, the tumor growth was remarkably inhibited, which was due to the effective antigen presentation on the tumor cells. Overall, the MMP9-responsive conjugates carrying foreign antigens might be a highly potent alternative to CTL-based cancer immunotherapeutics. |
