Development of Small Polymeric Nanocarriers for Systemic siRNA Delivery |

siRNA elicits the sequence-specific degradation of mRNA in the cytosol of mammalian cells, thereby highlighted as a potential therapeutic agent for intractable diseases, such as cancer. However, there are several biological hurdles for successful siRNA therapies; e.g., enzymatic degradation, rapid renal clearance, and inefficient cellular uptake of siRNA. To overcome these hurdles, we have elaborated block copolymers of PEG and cationic poly(amino acid) for construction of polyion complex (PIC) micelles. PIC micelles have siRNA-loaded PIC core and biologically inert PEG shell, protecting siRNA from enzymatic degradation and rapid renal clearance. Also, siRNA-loaded PIC micelles have a size of <50 nm, allowing efficient tumor accumulation through the leaky tumor vasculature. In this presentation, we introduce our recent polymer design strategies for construction of smart micellar nanocarriers and ultra small nanocarriers, directed toward enhanced tumor accumulation of siRNA. |
