In this study, we introduce a new technique to fabricate a sieve structure inside a microchannel via photopolymerization adopting UV-curable precursor solutions, and apply the structure for DNA purification. The precursor solution was prepared by mixing polyethylene glycol diacrylate (PEGDA) and methacrylate-based silane at an optimum ratio, and the mixture was selectively polymerized inside a microchannel. The microsieve can be formed selectively at a specified region of the microchannel. Preliminary experiments were performed to find an optimum mixing ratio of the two chemicals, and spatially-defined polymerization was examined. By using methacrylate-based silane reagent, we attempt to functionalize the terminal of the microsieve to perform nucleic acid purification. |
