Caprolactam is amonomer used for the synthesis of nylon-6. An intracellular biosensor for caprolactam canfacilitate high-throughput metabolic engineering of recombinant microbialstrains. Because of the mixed production of caprolactam and valerolactam inthe recombinant strain, a caprolactambiosensor should be highly specific for caprolactam.However, a highly specific caprolactamsensor has not been reported. Here, we developed an artificial riboswitch thatspecifically responds to caprolactam.This riboswitch was prepared using a coupled invitro-in vivo selection strategy with a heterogeneouspool of RNA aptamers obtained from invitro selectionto construct a riboswitch library used in invivo selection.The caprolactamriboswitch successfully discriminated caprolactam fromvalerolactam.This riboswitch enabled caprolactam-dependentcontrol of cell growth, which will be useful for improving caprolactamproduction and is a valuable tool for metabolic engineering. |
