Chemically-Conjugated Bone Morphogenetic Protein-2 and Vitamin D-TGF on porous PCL/BCP-Collagen scaffolds for enhance bone regeneration |

In this study, porous polycaprolactone (PCL)-biphasic calcium phosphate (BCP) was immobilized with Collagen, Bone morphology protein-2 (BMP-2) and VitaminD-TGF (VitD-TGF) successfully to improve the bone regeneration. The surface of PCL/BCP scaffolds was aminolyzed by 1,6-hexamethylenediamine to introduce amino groups onto their surface. Then Collagen, BMP-2 and VitD-TGF were further immobilized on the aminolyzed porous PCL/BCP via EDC/NHS crosslinking agent. The pore size of scaffolds was 200-300 µm which was suitable for bone in-growth. After immobilization, the pre-osteoblast cell viability and proliferation were improved remarkably confirmed by MTT assay, Live-dead assay and immunofluorescence. The in vivo study will be carried out by implanting the porous PCL/BCP-Collagen, PCL/BCP-Collagen-BMP-2 and PCL/BCP-Collagen-VitD –TGF to the skulls of rats. The present results provided more evidence that functionalizing PCL/BCP with BMP-2 and VitD-TGF may be a feasible way to improve the interaction between cells and also , PCL/BCP-Collagen-BMP-2 and PCL/BCP-Collagen-VitD –TGF scaffolds could be a potential candidate for bone regeneration. |
