Effects of recombinant human bone morphogenetic protein-2 on in vivo bone regeneration of fibrous BCP-PCL/GEL composite |

To confirm effect of recombinant human bone morphogenetic protein-2(rhBMP-2) for bone regeneration, rhBMP-2 loaded biphasic calcium phosphate(BCP)-polycaprolactone(PCL)/gelain(GEL) composite fibrous scaffolds were fabricated successfully using electrospinning method. Compared with BCP-PCL/GEL scaffold, the rhBMP-2 loaded BCP-PCL/GEL scaffold shows improved biocompatibility, cell proliferation and cell adhesion behavior evaluated by in-vitro studies. The BCP-PCL/GEL scaffolds with rhBMP-2 or without were implanted in rat skull defects for 4 and 8 weeks to evaluate the detailed biological response physiological condition. In vivo bone regeneration was compared by evaluating the degrees of new bone formations using micro-CT scanning, and histological staining using hematoxylin & eosine / Masson’s trichrome staining method. The defect implanted with electrospun mat containing rhBMP-2 demonstrated highest amount of bone tissue regeneration among the samples. This result suggests that rhBMP-2 loaded BCP–PCL/GEL membranes could be considered as potential bone tissue engineering applications. |
